Candidate-gene identification past and present. In the past, the emphasis was on using
linkage analysis to direct positional-cloning efforts, and on the application of our
limited understanding of disease pathogenesis to select biologically relevant candidate
genes. Now, additional techniques are providing new routes to the identification of
disease-susceptibility genes. See text for further details.
