Lentiviral RNAi library screen of human kinases identifies regulators of cancer cell
growth. (a) Target genes covered by the shRNA library were classified according to gene function
using Gene Ontology groups [33]. (b) Two human sarcoma cell lines, HT1080 and U-2-OS cells, were infected with lentiviruses
containing shRNAs targeting human kinases in 384-well format. Genes whose knockdown
inhibits growth of either cell line by more than 25% compared to control with statistical
significance (P < 0.01) were considered as hits. This diagram shows the number of hits specific to
each cell line and common to both cell lines. (c) Median value of percent growth inhibition (%GI) from triplicate results in each cell
line. The top seven hits were selected based on the summed value of %GI in both cell
lines.
